Our group develops computational techniques that illuminate the cellular organization of complex tissues. We apply these techniques, together with high-throughput sequencing, single-cell genomics, and experimental approaches, to understand how tumors develop, progress, and respond to therapy. As part of this focus, we develop new algorithms to resolve cell states and multicellular communities, tumor developmental hierarchies, and single-cell spatial relationships from genomic profiles of clinical biospecimens. We have a particular interest in developing algorithms that can probe the molecular and cellular basis of tissue development, both in space and in time, since stem cell regulatory programs influence how tumors grow, evade clearance by the immune system, resist therapy, and disseminate throughout the body. Key findings from our computational efforts are further explored experimentally, both in our lab and through collaboration, with the ultimate goal of translating promising findings into the clinic.
